I'm trying to work out a mystery 375 chambering for a rifle while I await a chamber cast.
It's a break open rifle that started out as a 375 Winchester and now it has been rechambered. Rough measurements leave me to believe it may be a 375-348 improved or a 375 Alaskan. I found a drawing on the later and some of the measurements work out.
If it turns out to be one of these how difficult is it to make cases and the positives or negatives of loading for a cartridge like this?

Sourcing dies, brass forming and reliable loading data are your biggest issues. If it does turn out to be something like a 375 Alaskan, it will be quite a useful cartridge.

Once you have a set of dies it should be no more difficult to load than any other wildcat. You will have to form your own cases and some of the more complex designs require several operations before you have useable brass.

As you aren't restricted to flat nose bullets in a single shot you have a wide range to select from in 375 cal.
