Or
NXS 5.5-22x50 illuminated MOAR

I ain't following why the twist makes a difference as to what scope to use.
Nightforce
I have nightforce on my fast twist 22-250. SHV 3-10

Given the two scope options it doesn't, but that notwithstanding—effective range.
I own both brands and the more precise the adjustments have to be, the more Nightforce becomes the pick.
Well think about how long Nightforce has been here, then ask yourself if any good shooting took place before that.
Mind you i own both brands and find they both perform very well.
As for prairie dogs, ive shot lots of them with a 22x250 AI which is my favorite gun to use on them.
I have a Sightron scope on that which works well also.
When i arrive at the spot im going to hunt that day i will asses where most of the shooting will be as for distance with that particular gun. I will zero on the first dog at that distance, and from then on i dont do any more dialing, just hold up or down and left or right depending on wind.
Two reasons for that, dialing for each one takes too much time is one, and unnecessary is the other.
Anyway as anybody who has hunted them knows, close misses can be very entertaining also.
Shots longer than what that gun is capable of with any degree of consistency would be a different thing.
But even then Nightforce quality and cost isnt necessary.

Its a rhetorical question anyway...
Hmmm.....I'd not pick either of those. But it's your money. Go for the NF.

This would be my choice

Night force
Not sure if the OP from Wisco is trying to ask a legitimate question or stir the fire pot ????????????!!!!
I probably wouldn't do either. If I put together a very accurate .22-250 for long range work, it would probably improved or I'd go with .22 Creedmoor, and for the same money as the 2 scopes you mention, I'd do something else ... Nightforce 12-42X benchrest.

NXS 5.5-22x50 illuminated MOAR-T
A couple things, Leupold has discontinued the Varmint Hunter Reticle both illuminated and non throughout their entire line quite some time ago. IMO an illuminated reticle is not really necessary for prairie dogs and ground hogs, they normally are best hunted when the sun is out.
I have several 1-8" twist 22-250 and 22-250AI's most wear a Leupold VX-3 6.5-20x 50 or a 4.5-14x40.
The NXS is a great scope but more than 10oz heavier if weight is an issue.
