Option 1:
Engage posi-lok for every shot. Try and use consistent torque every time.
Option 2:
Leave Posi-Lok disengaged while sighting in. When sighted in attach a good optical bore sighter (like the bushnell professional) and note where the cross hairs are on the grid. Engage posi-lok and see where the cross hairs move too. Disengage posi-lok and adjust scope to compensate for the movement. Re-egage posi-lok and check. Repeat till it is in the same spot as previously noted with the posi-lok engaged. Fire some shots to make sure the POI is the same as before.
Just two ideas I came up with.

Your option #2 will not work.
Option #1 is pretty much right on.
The correct wayt to use the Posi loc is to shoot the rifle, disengage the loc, make adjustments, then lock it down, and repeat as needed.
There is an Option #3 which is to disengage the Loc and leave it disengaged, which will make your scope just like anyother Burris signature. There is a very strong 15 lb retention spring that holds the inner scope tube in place and the industry standard on scopes of that price range is 7 1/2 lbs.
No doubt that the Burris Posi lock will take some recoil of some very heavy rifles and has a super tough scope to boot. I had two friends in S. Ca. that would not take any other scope to Africa other than the Burris with the posi loc. I have used probably half a dozen of them when I was coyote hunting, having them bounce around all over the front seat of the truck and back of the truck,and carried in scabbord on horses and mules...they are tough as hell. Those wash board roads in Az will "field test" any good scope to death.

I haven't tried either yet, but I was wondering why you think #2 won't work? I will be scoping in the .45-90 this weekend and was thinking I'll test both.

#2 will not work, try it and you will pull your hair out trying to get it to work. This is something that I have had hands on experience with. Follow my directions and you will have a great day, do otherwise, and you will not be a happy camper.
After trying #2 without success, I read the directions that came with the Posi Loc scope...Duhhhh.....
