Not a wheel gun, but I used a 375 JDJ with a Hornady 270 gr bullet in a Contender. I was comfortable out to 300 yds with a rest but it turned out to be a 30 yd shot. Bang - flop
Deer antelope and pigs have fallen to 243s, 44s, 357 Max and a 445 Super mag. The 2 handguns that don't have blood on them are an XP in 338 Federal and a newly acquired Striker that will become a 6.5 CM.
